ABSTRACT

Objective@#To investigate the curative effect of local application of CpG-oligodeoxynucleotide (CpG-ODN) combined with 4-1BB monoclonal antibody in hepatoma-bearing mice, and to evaluate the effect of 4-1BB monoclonal antibody on CpG-ODN immunotherapy.@*Methods@#H22 single cell suspension was injected subcutaneously into the axilla and four limbs of the BALB/c male mice to establish a tumor-bearing mice model. After 7 days, 30 mice with corresponding tumor-bearing volume were screened and randomly divided into model control group, CpG group and CpG+4-1BB group, and the drug was injected into the tumors of left lower extremity. The same batch of normal mice was selected as normal control group. Survival of mice was recorded. Tumor-bearing volume and organ index were calculated. Serum levels of interleukin (IL) - 12 and interferon (IFN) gamma and spleen CD8+T lymphocyte ratio were measured. The measurement data were analyzed by analysis of variance. The survival rate of each group of mice was analyzed by log-rank test.@*Results@#Mice in the model control group with tumor-bearing volume had a sustained growth before the execution. CpG group and the CpG+4-1BB group [(976.08 ± 29.55) mm3, (47.25 ± 0.93) mm3)] tumor-bearing volume was decreased than model group [(1 336.52 ± 39.40) mm3] (F = 5 329.273, P < 0.05). CpG+4-1BB group distant tumor-bearing volume [(611.83 ± 113.02) mm3] was decreased than model group and CpG group [(1 406.62 ± 51.09) mm3, (1 380.01 ± 51.44) mm3] (F = 247.160, P < 0.05), but there was no significant difference between the CpG group and the model group (P > 0.05). Serum IL-12 concentration (23.90 ± 2.33 pg/ml), IFN-γ concentration (103.02 ± 6.10 pg/ml) and spleen CD8+T cell ratio (4.54 ± 0.62%) in the model group were lower than those in the normal group (P < 0.05). Serum IL-12 concentration in CpG group and CpG+4-1BB group (29.21 ± 2.23 pg/ml, 37.04 ± 1.49 pg/ml), IFN-γ concentration (116.12 ± 4.08 pg/ml, 138.65 ± 1.72 pg/ml), CD8+T cell ratio (6.65 ± 0.64%, 12.73 ± 0.88%) were higher than the model group, while CpG+4-1BB group was higher than the CpG group (P < 0.05). The survival rate of CpG+4-1BB group was higher than that of model group and CpG group (χ2 = 25.544, P < 0.05), but there was no significant difference between CpG group and model group (P > 0.05). There was no significant difference in organ index between the four groups (P > 0.05).@*Conclusion@#4-1BB monoclonal antibody combined with CpG-ODN therapy can shrink hepatoma-bearing capacity, inhibit the growth of distant tumors and significantly prolong the survival time of mice.

ABSTRACT

Non-alcoholic fatty liver disease (NAFLD) is a common and poorly understood liver disease whose incidence is globally rising.The prevalence of NAFLD is 25%-30%,and its main pathological change is the pathological accumulation of lipids in the liver.At present,research has confirmed that gut microbiota play a crucial role in the development of NAFLD.Gut microbiota may influence the NAFLD pathogenesis by increasing the inflammation in liver,altering the intestinal permeability,and affecting energy and substance matabolisms.This article reviews the roles of gut microbiota in the pathogenesis and treatment of NAFLD.

ABSTRACT

Objective: To explore the protective mechanism of resvertrol (RSV) suppressing the rapid electrical stimulation incurred oxidative stress injury in neonatal rats cardiomyocytes. Methods: The neonatal rats cardiac ifbroblasts and myocytes were isolated by double enzyme digestion and differential adhesion method, and the cardiomyocytes were randomly divided into 5 groups:①Control (CTR) group,②Rapid electrical stimulation (RES) group,③RES+APO group, cells were pretreated with NADPH oxidase inhibitor APO,④RES+RSV group,⑤RES+AIP group, cells were pretreated with CaMKII inhibitor AIP. In order to conifrm whether RSV protection was via MsrA-OX-CaMKⅡpathway, the cells were further divided into another 3 groups:①DMSO control group,②RSV+DMSO group,③RES+ RSV+DMSO group. The best dose of RSV was measured with Kit-8 by cardiomyocytes surviving condition, the optimal electrical stimulation time was detected with ELISA by Ang II level in conditioned medium. The level of reactive oxygen species (ROS) in cardiomyocytes was detected by flow cytometry, the protein expressions of MsrA, Nox4, Nox2, P22phox, OX-CaMK II and apoptosis related cleaved caspase-3 were observed by Western blot analysis.Results:①Compared with CTR group, RES group showed increased AngII secretion with increased protein expressions of Nox4, Nox2, P22phox, OX-CaMK II and cleaved caspase-3.②Compared with RES group, the RES+APO, RES+RSV, RES+AIP groups had decreased ROS level, the ROS was even lower in RES+RSV group.③Compared with RES group, the RES+APO, RES+RSV groups presented decreased protein expressions of Nox4, Nox2, P22phox, OX-CaMK II and cleaved caspase-3, while RES+AIP group only had decreased Nox2, OX-CaMK II and cleaved caspase-3.④Compared with DMSO control group, RES+ RSV+DMSO group had the lower level of cleaved caspase-3 expression. Conclusion: RSV has protective effect on rapid electrical stimulation incurred oxidative stress injury in neonatal rats cardiomyocytes, which might be via NADPH oxidase with the increased MsrA expression .

ABSTRACT

Objective: To explore the effect of resveratrol (Res) on angiogenesis of human umbilical vein endothelial cells (HUVEC) with the possible mechanisms in vitro. Methods: The HUVECs were cultured in 6 groups.①Control group, HUVEC were cultured with high glucose in DMEM,②Res group, the cell were cultured with Res at different concentrations,③Res with PI3K blocker LY294002 (Res+Blocker 1) group, ④Blocker 1 group, HUVEC were cultured with LY294002 alone, ⑤Res with eNOS blocker L-NAME (Res +Blocker 2) group and ⑥Blocker 2 group. The effect of Res on HUVEC proliferation was detected by CCK-8 kit, the protein expressions of p-Akt, p-eNOS were examined by Westin blot analysis, nitric oxide (NO) level was measured by nitrate reduction method and the endothelial cell migration was assayed by transwell chamber method. Results: ① Compared with Control group, HUVEC proliferation increased in Res (1, 5μmol/L ) group, P0.05.④Compared with Control group, the cell migration and tubing formation were higher in Res (5μmol/L) group, P Conclusion: Res could up-regulate NO level via activating PI3-K/Akt/eNOS signaling and therefore, improving the proliferation, migration and tubing formation of HUVEC in vitro.

To better accomplish the duty of medical support mission, the problems of medical equipment and material store house management in the field medical station are discussed. Some targeted suggestions are introduced from daily management and the use of equipment respectively. It can be used as reference for other military hospitals to accomplish different medical support tasks.

ABSTRACT

Objective To develop a database management system specially applied for medical equipment maintenance administration, Methods Under PowerBuiider/SQL Server development environment, the C/S mode system structure was confirmed and the system function modules were designed. Results The application of C/S mode structure was coincident with the division-cooperation work flow in large and medium hospitals, and the design of function modules met the demand of information collection and utilization of medical equipment maintenance.Conclusion The database management system can preserve the information of medical equipment maintenance effectively and facilitate its usage. Moreover, many statistical indexes about the maintenance administration are available in this system.

ABSTRACT

Objective To investigate the changes between type 2 diabetes mellitus(T2DM) with peripheral vessel disease and osteoporosis. Methods A total of 100 cases T2DM patients were divided into non-peripheral vessel disease (A group) and peripheral vessel disease (B group). B-mode ultrasonography to measure the intima-media thickness(IMT) of carotid artery,dual energy X-ray absorptiometry (DEXA) to measured bone mineral density(BMD) and VS-1000 arteriosclerosis diagnometer to measure brachi-ankle pulse wave velocity (baPWV) were used in 100 T2DM patients. The clinical data and BMD were compared between two groups. Results Compared with normal group,the BMD in T2DM patients significantly decreased.Compared with T2DM patients without peripheral vessel disease,the BMD decreased in T2DM patients with peripheral vessel disease. Conclusions Atherosclerosis in T2DM with peripheral vessel disease is probably related to osteoporosis, and the relationship between them is worth further investigation.

Results Data exchange between client and server was realized. Meanwhile, the designed function modules achieved normal operation. Conclusion The system structure and functions meet the requirements of working practice in medical engineering department.

Objective To perform multi -condition combination query in the browse window based on PowerBuilder datawindow technology. Methods After the interested combination of query conditions specified on certain columns in datawindow was constructed by if...then statement, the setfilter () and filter () function was used to retrieve and display data which met the filter criteria. Results A flexible query object was incorporated into the data browsing window. Conclusion The datawindow-based query is suitable for situations where batch of data are filtered against not so many columns.

ABSTRACT

Objective To access images of medical equipments in database management system. Methods Images in SQL Server2000 can be stored inside database tables directly as the binary content of a Blob field. Under the PowerBuilder development environment, SelectBlob and UpdateBlob are used to perform the data exchange between the Blob field and a Blob variable defined in Powerscript,then image contained in the Blob variable can be constructed in a picture control available in PowerBuilder. Results It is convenient and expeditious to insert, update and present images in this PowerBuilder application. Conclusion Processing images in the medical equipments database management system extends its functions and has a significant applied value.

ABSTRACT

Objective To evaluate the formation of coronary collateral circulation with intracoronary Doppler ultrasound in pigs after acute occlusion and the influence of nitroglycerin and phentolamine on it.Methods The blood flow spectrun was detected in 14 pigs using intracoronary Doppler ultrasound before and after acute occlusion of the left circumflex artery (LCX) and its response to nitroglycerin and phentolamine.The average peak velocity (APV) and the diastolic systolic velocity ratio (DSVR) were recorded. Results The APV of the LCX decreased significantly at 10 min after acute occlusion (0.97?1.05 vs 25.78?4.36 cm/s,P0.05). Conclusion Intracoronary Doppler ultrasound can be used to evaluate coronary collateral circulation. The formation collateral circulation was insufficient after acute occlusion of coronary artery in pigs and nitroglycerin or phentolamine can improve the blood flow.
